Carboxypeptidase Cathepsin X Defines a Multifunctional Role of Gamma- Enolase in Cancer

Gamma-enolase enzymatic activity is involved in glycolysis, a prevalent process cancer cell metabolism. Additionally, gamma-enolase has pro-survival function, exhibited through the active site at C-terminal end of molecule. This regulated by cysteine peptidase cathepsin X, which cleaves two amino acids gamma-enolase. In clinical practice, determination as tumour marker does not differ between total, uncleaved and C-terminally cleaved forms. However, levels alone may provide additional information. this study we analysed C- terminally total lines sera from 255 patients with colorectal (CRC) western blot, immunoprecipitation, activity, ELISAs ECLIA. Results show that gamma-enolase, rather than gamma- enolase, exhibits different cells, being highest those, derived metastatic sites or highly invasive tumours. secreted into extracellular space predominantly an form were congruent to those within cells. Furthermore, cells are inversely related X protein level its activity. Uncleaved also predominant CRC. Both forms exhibit similar stage dependent distribution, slightly elevated IV patients. Higher significantly shorter survival support evidence function cancer. Future studies should focus on analysis samples, relations indicators disease progression.
